If you work in process chemistry or sourcing, you’ve probably seen this one pop up on RFQs: 2,2-Dimethylbutyryl chloride (CAS 5856-77-9). It’s not as ubiquitous as pivaloyl chloride, but it carves out a nice niche where a slightly longer, still-bulky acyl fragment is desired. And yes, it’s moisture sensitive and a bit of a diva in the drum—more on storage in a second. We’re seeing steady demand from custom synthesis groups and agro innovators who need sterically demanding acylations. Compared with lighter acyl chlorides, this one tends to dial up hydrolytic stability in downstream esters/amides. Some customers say it’s become their “go-to” blocker when they want a bulkier cap but not the full pivaloyl footprint—interesting middle ground, I’d say. Materials: 2,2-dimethylbutanoic acid, thionyl chloride or oxalyl chloride; catalytic DMF; dry solvent as needed. Method: controlled addition, moisture exclusion, HCl/SO2 venting, and then a careful distillation (reduced pressure) for a clean cut. Testing: GC assay and related substances; KF moisture; color (APHA); identity by IR and 1H/13C NMR; sometimes chloride titration. Standards used commonly include ASTM E203 and D1209, with GMP-aligned documentation (ICH Q7) when supplied as an API intermediate. Service life: unopened drums often hold 18–24 months at 2–8°C under nitrogen; once opened, many teams aim to finish within weeks—water is the enemy here. Advantages include strong steric bulk without going fully neopentyl, relatively neat distillation behavior, and—surprisingly—good batch-to-batch reproducibility reported by several buyers. Custom GC thresholds, low-acid grades, or tighter APHA limits are common requests. Some projects also ask for additional NMR trace reporting and residual solvent panels per ISO/IEC 17025-tested methods. To be honest, it’s worth asking for a mock CoA early. GHS/CLP: corrosive; lachrymator; reacts with moisture to release HCl. Use dry nitrogen, acid-resistant PPE, and vented caps. Check SDS and local regulations before scale-up.
